Researchers have found that the spread of a dangerous bacteria that can be fatal in up to 45% of people infected in sub-Sahara Africa may have been instigated by the emergence and spread of HIV in Africa. The study also found that one of the major contributing factors for the successful spread of the disease -- non-Typhoidal Salmonella (iNTS) -- was the acquisition of genes that afford resistance to several front line drugs used to treat blood-borne infection such as iNTS.
A new study out September 30 reveals that the emergence and spread of a rapidly evolving invasive intestinal disease, that has a significant mortality rate (up to 45%) in infected people in sub-Saharan Africa, seems to have been potentiated by the HIV epidemic in Africa.
The team found that invasive non-Typhoidal Salmonella (iNTS) disease is caused by a new form of the bacteria Salmonella Typhimurium that has spread from two different focal hubs in Southern and Central Africa beginning 52 and 35 years ago, respectively. They also found that one of the major contributing factors for the successful spread of iNTS was the acquisition of genes that afford resistance to several front line drugs used to treat blood-borne infection such as iNTS.
iNTS is a blood-borne infection that kills approximately one of four people in sub-Saharan Africa who catch it. Yet, in the rest of the world, NTS is a leading cause of acute inflammatory diarrhea that is self-limiting and tends to be fatal in less than 1 per cent of people infected. The disease is more severe in sub-Saharan Africa than the rest of the world because of factors such as malnutrition, co-infection with malaria or HIV and potentially the novel genotype of the Salmonella bacteria.
"The immune system susceptibility provided by HIV, malaria and malnutrition at a young age, may provide a population in sub-Saharan Africa that is large enough for this detrimental pathogen to enter, adapt, circulate and thrive," says Chinyere Okoro, joint first author from the Wellcome Trust Sanger Institute. "We used whole genome sequencing to define a novel lineage of Salmonella Typhimurium that is causing a previously unrecognised epidemic across the region. Its genetic makeup is evolving into a more typhoid like bacteria, able to efficiently spread around the human body"
From sequenced samples, the team created a phylogenetic or 'family tree', depicting the pathogen's evolution, dating when each sample first emerged and overlaying this with geographical information about where these samples came from. They found that this invasive disease comprises of two very closely related waves; the first wave originated from a possible south-eastern hub, about 52 years ago and the second originated about 35 years ago, possibly from the Congo Basin.
"The HIV epidemic in sub-Saharan Africa is thought to have begun in a central region and underwent expansion eastwards, a strikingly similar dynamic to that observed for second iNTS wave," says Dr Robert Kingsley, joint first author from the Wellcome Trust Sanger Institute. "Our findings suggest the current epidemic of iNTS and its transmission across sub-Saharan Africa may have been potentiated by an increase in the critical population of susceptible, immune-compromised people."
The team identified that the vast majority of samples from the second wave of iNTS contains a gene that makes them resistant to chloramphenicol, a frontline antibiotic in the treatment of Salmonella. This gene was not present in the samples from the first wave of iNTS. This observation suggests that iNTS acquired this gene early on in the evolution of the second wave, probably around the time of its spread from the Congo basin.
"Because it acquired resistance to chloramphenicol, this pathogen has much greater opportunity to survive and spread across the region," says Professor Gordon Dougan, lead author from the Wellcome Trust Sanger Institute. "This is the first time that the power of whole-genome sequencing has been used to track the spread of iNTS. Our research highlights the power this approach has to monitor the emergence and spread of dangerous pathogens both locally and globally over time."

The Casio XJ-H2650 isn't Casio's brightest projector, but it's part of Casio's Pro Series, which is its brightest line of projectors. Casio rates the XJ-H2650 at 3500 lumens, a bit less bright than the 4,000 lumen Casio XJ-H1750 Pro Series that I reviewed earlier this year. However, the XJ-H1750 offers a native XGA (1,024 by 768) resolution.
The only other Pro Series model with WXGA resolution is the XJ-H2650's near twin, the Casio XJ-H2600 ($2,000 street). Casio says that the two models are identical except for the addition of USB A, USB B, and LAN ports to the XJ-H2650, along with features that depend on those ports. Most notably, the XJ-H2650's USB A port lets you plug in a supplied Wi-Fi adaptor so you can connect directly with, and send data images from, Windows and Mac computers as well as most Android, iOS, and Windows smartphones and tablets.
The extra ports also let you plug in a memory key to read JPG and a few other file formats directly, control the projector over a network, send data images over a network, and add interactivity with an interactive pen and software option ($260 street). However, the interactive feature is hard to recommend, since the projector's standard-throw lens would make it hard to interact with the image without casting shadows that would get in the way. All other features on the XJ-H2600 and XJ-H2650 should be identical, so the rest of the comments in this review should apply to both.
Basics, Setup, and Connections
Like all current Casio projectors, the XJ-H2650 is built around Casio's hybrid light source, consisting of LEDs and a laser paired with a DLP chip. The LEDs produce the red and blue primaries directly. The laser shines on a phosphor element to produce green.
Not too surprisingly, given that both projectors are part of the same Pro Series, the XJ-H2650 shares a lot of its basic features with the XJ-H1750. In particular, it weighs almost the same, at 15.6 pounds, which makes it most appropriate for permanent installation or room to room portability on a cart.
Setup is standard, with a manual focus and a manual 1.2x zoom, which gives you some welcome flexibility in how far you can put the projector from the screen for a given size image. In addition to the LAN and USB ports I've already mentioned, connection choices include the usual HDMI for a computer or video source, VGA for computer or component video, and both S-video and composite video inputs.
Brightness and Eco Modes
In my tests, the XJ-H2650 was easily bright enough in its brightest mode for a 78-inch wide (92-inch diagonal) image to stand up to the typical level of ambient light in an office or classroom setting. Even better, it was bright enough at that size so you could easily take advantage of its eco modes to minimize power use and save on electricity.
Most projectors offer one bright mode and one eco mode that lets you lower brightness and power consumption. Beyond that, they offer color presets that can also affect brightness, but are, in theory, meant to adjust color.
The XJ-H2650 gives you more flexibility for brightness and power use, with two non-eco modes, which I measured at 340 watts and 290 watts, plus five eco modes, with a measured range of 260 to 115 watts. With multiple eco modes, you have the freedom to pick the best color setting first, and then choose the least bright mode with the brightness you need, instead of using the color presets to adjust brightness. Also worth mention is that the only projector I've seen with more eco-mode levels is the laser-based BenQ LX60ST that I recently reviewed.

ScienceDaily (Sep. 28, 2012) ? Researchers at The Ohio State University have discovered a new class of treatment against methicillin-resistant Staphylococcus aureus (MRSA) as well as evidence of a growing need to quickly genotype individual strains of the organism most commonly referred to as the "superbug."
The two separate studies were funded by the Ohio State Center for Clinical and Translational Science (CCTS) with a goal of increasing the MRSA knowledge-base from both a basic science "bench" perspective, as well as using real-time data from infected communities to determine how MRSA spreads.
"The public is most familiar with the dramatic progression of skin infections caused by MRSA, but MRSA is responsible for a range of difficult to treat illnesses," noted Dr. Kurt B. Stevenson, professor of Infectious Diseases at the Ohio State College of Medicine, and primary investigator of the CCTS-funded study following the transmission of MRSA infections in communities. "While we've seen a decrease in the number of MRSA cases, identifying new agents and tracking methods will be critical to stopping these infections before they can start."
Cancer treatment search leads to potential MRSA killer, immune booster
In 2002, Ching Shih Chen, professor of medicinal chemistry at Ohio State College of Pharmacy, and a team of researchers were creating a library of anti-cancer agents built around the scaffold of the molecules of celecoxib, a popular arthritis treatment in a family of drugs known as cyclooxygenase-2 (COX-2) inhibitors. This effort yielded OSU-03012 (AR12), a compound that is currently in a Phase I clinical trial as anticancer agent at the OSU Comprehensive Cancer Center -- Arthur G. James Cancer Hospital and Richard J. Solove Research Institute.
After observing how the substance acted within breast cancer cells, Hao-Chieh Chiu, a then postdoctoral researcher in Chen's lab, realized that the derivatives were suppressing a mechanism that bacteria also use to take over their host cells. Chiu decided, with the support of Dr. Chen, to focus his research on testing the COX-2 compound library against a variety of bacteria.
"When the compounds showed anti-bacterial activity against Salmonella and Francisella, we began testing efficacy against a variety of pathogenic bacteria, including Staphylococcus aureus, Enterococcus, and Streptococcus," said Chiu, who is an assistant professor at the Department of Clinical Laboratory Sciences and Medical Biotechnology at National Taiwan University. "It became clear that these celecoxib analogues had a unique anti-bacterial activity, and they appeared to be most potent against Staph aureus and other MRSA strains."
The researchers narrowed the library down to a single agent (dubbed "compound 46") and moved to testing in MRSA-infected mice. Published in the August issue of Bioorganic and Medicinal Chemistry, the authors report that an intraperitoneal administration of compound 46 resulted in increased survival in MRSA-infected mice versus untreated mice. "It was particularly gratifying to see that these compounds, originally designed as anticancer agents, work as a novel class of anti-bacterial agents based on the same principle in bacterial cells," said Chen.
The researchers are hopeful that this early work will ultimately provide insights on the development of a treatment for antibiotic resistant infectious diseases. The team is already working with scientists at the Ohio State Center for Microbial Interface Biology, led by Dr. Larry Schlesinger, to use this technology to develop novel agents against tuberculosis, another public health threat facing similar issues with drug resistance.

The team, funded by the Centers for Disease Control and Prevention (CDC), used diverse methods from geographic analysis to molecular genotyping to track more than 1,000 MRSA cases from the Wexner Medical Center and community hospitals across Ohio. Experts say using a variety of tracking methods is essential to stopping infections before they start.
"With data from different sources, we've markedly improved our understanding of how MRSA is acquired and then spread among healthcare facilities. For example, we identified a very rare strain in the US, ST-239, which originated in Asia, spread to hospitals in Western Europe, and was introduced to Ohio sometime in the past two decades. It's that level of knowledge that will help us change the course of transmission," said Stevenson, whose research on ST-239 and its presence in healthcare facilities was published online ahead of print in the October issue of Emergent Infectious Diseases.
Using funding from the CCTS, investigators further applied diverse tracking methods to bloodstream isolates from hospitals in Franklin County, Ohio, as well as skin and soft tissue infections among patients receiving primary care in a variety of settings. The results not only demonstrated the value of rapid molecular typing in examining the distribution and transmission of individual MRSA strains, but showed that particular strains tended to cluster in specific places.
"These studies have demonstrated that specific molecular types of MRSA are linked to specific types of infections, or even specific settings. For instance, there are strains that tend to colonize catheters, strains that are more commonly found in nursing homes," said Shu-hua Wang, assistant professor of Infectious Diseases, who worked with Stevenson as part of a mentorship grant from the CCTS. "As we understand why certain MRSA strains behave as they do, more targeted interventions for prevention and treatment can be tested."
Stevenson is hopeful that technology will someday provide a quick and inexpensive on-site genomic analysis of MRSA. In anticipation of that day, the team is using the "roadmap" data to create a MRSA molecular library that provides a detailed background on individual strains, including its drug resistance, weaknesses, and most likely source of transmission. "We're envisioning a future where every patient admitted into a hospital will get a rapid, strain-specific MRSA test and within minutes, a doctor will know exactly what protocol to follow to stop the patient from getting sick, and stop the bacteria from spreading," said Stevenson.

SANAA, Yemen (AP) ? Yemen's president said Tuesday he would talk to his country's branch of the al-Qaida militant network if it were to give up its weapons, abandon its extremist ideas and stop sheltering foreign fighters.
The televised statement by President Abed Rabbo Mansour Hadi appeared aimed at showing his readiness to use any means to reduce the political violence in his country, home to the most world's most active branch of the terror network. Al-Qaida however has shown little indication that it would want to pursue a dialogue, especially under Hadi's conditions.
The militants had taken advantage of Yemen's popular uprising last year to seize much of the country's south. This summer, backed by U.S. advisers and drones, Hadi's military launched an offensive to reclaim this territory, going after al-Qaida's strongholds and hideouts. The groups' No.2 was killed earlier this month in an airstrike.
The militants in turn have struck back, targeted security officials for assassinations and staging deadly bombings.
Hadi's remarks were in line with the image he tries to project, of a leader serious about bringing stability to his country either by force or by negotiation.
He said he was pressured by unidentified local "mediators" to accept the idea of dialogue with the militants. The group has often found shelter among tribal leaders.
"I say, despite the heavy blood that was shed and the homes which were destroyed, and people who lost their homes, a dialogue can be started on condition that the al-Qaida agrees publicly to hand over its weapons, declares its repentance from its extremist ideas that have nothing to do with Islam and gives up sheltering armed groups from outside Yemen," he said. "Then this would be opening the door for dialogue."
He added, however, that "mediators go and never come back," indicating skepticism that al-Qaida would ever take up the offer.
Al-Qaida in the Arabian Peninsula has planned attacks against targets in both Yemen and the United States., including the failed attempt by an al-Qaida operative to detonate a bomb in his underwear aboard a flight bound for Detroit on Christmas Day, 2009.
In May, nearly 100 Yemeni soldiers were killed when a suicide bomber blew himself up at a parade rehearsal. Al-Qaida said at the time it was targeting the defense minister, who was the target of at least six assassination attempts. At least 13 people were killed in the latest attempt against the minister two weeks ago in Sanaa.
Hadi's predecessor Ali Abdullah Saleh was long accused by critics of not fighting al-Qaida wholeheartedly. Saleh stepped down in February in a power transfer deal backed by the U.S., but Hadi and his supporters complain the former leader undermines security using relatives and loyalists still in key positions.
On Tuesday, Hadi vowed to continue to free the country from "burden of the past," an apparent reference to his predecessor's alleged meddling. "All I promise is that I will not be silent or objective before whatever curtails the path to change," he said.
The impoverished Arabian Peninsula nation's economy has been hard hit by the upheaval.
On Tuesday, an explosion struck a gas pipeline in southern Yemen, temporarily halting the flow of gas although exports were not affected, a government official said. The pipeline has repeatedly been targeted by groups suspected to be affiliated with al-Qaida.
An oil ministry official said repair work has begun in the Liquefied Natural Gas Co. pipeline. He spoke on condition of anonymity because he was not authorized to speak to reporters.
The bombing in Shabwa province comes on the heels of a Monday attack against an oil pipeline nearby.
A statement by the gas company said nobody was hurt in Tuesday's explosion. Yemen LNG Co. was created in 1995 ? the country's first natural gas liquefaction consortium. Its major shareholders include French oil company Total SA and Dallas-based Hunt Oil Co. It also is backed with Asian, U.S. and Yemeni government funding.

Mansor called 9-1-1 on the afternoon of June 12 last year, reporting that his son, 11-week-old Bryan, wasn?t breathing. Mansor told sheriff?s deputies he went online for advice when the baby struggled to breathe. About 15 minutes passed, he said, before he called 9-1-1.?
Bryan died the following day of abusive head trauma, doctors determined. Mansor is charged with Bryan?s death and the abuse of Bryan?s twin brother, Ethan.?
Detectives served a search warrant at the residence on Sutton Place and seized Mansor?s computers to verify his statement.?
Computer forensics analyst Justin Lazenby testified at trial Thursday about evidence found on the computers.?
Lazenby testified that Internet search terms on the computer included ?11 week old baby pulse no breathing,? ?abuse newborn symptoms,? ?how do I stop abusing my baby? and ?abuser therapy.??
Defense attorney Russell Barnett filed a pre-trial motion to suppress the evidence, arguing that the search of Mansor?s computers was unconstitutional. The motion said the lack of specificity in the search warrant yielded a broad, exploratory search that invaded Mansor?s privacy. Barnett argued police lacked probable cause for delving into so much of Mansor?s personal digital information.?
Judge Don Letourneau denied the motion, and Barnett objected Thursday to Lazenby?s testimony about the contents of Mansor?s Internet history on the basis of relevance.?
The state agreed not to ask Lazenby questions about a game downloaded on the computer called ?Candyvan,? which involves the player abusing children. However, jurors will receive Lazenby?s report, which mentions the game, in evidence.?
According to the report, the game is a ?simple DOS-based program where the protagonist drives around in a van/bus/ice-cream truck, picks up 10 children, and abuses them (physically and/or sexually) while trying to avoid attention and the police.? Certain player actions correspond in the game to words such as, ?You throw the kid to the ground and kick until there?s a pool of blood on the floor.??
Barnett withdrew his objection after prosecutors said they would not question Lazenby about the game.?

DUBAI (Reuters) - A senior Iranian lawmaker accused the head of the United Nations' nuclear watchdog on Sunday of passing confidential information about Iran's nuclear activities to Israel.
In the latest sign of strained relations with the International Atomic Energy Agency, Javad Jahangirzadeh, a member of parliament's presiding board, said IAEA chief Yukiya Amano would be to blame if Iran reduced its ties with the body.
"Amano's repeated trips to Tel Aviv and asking the Israeli officials' views about Iran's nuclear activities indicates that Iran's nuclear information has been disclosed to the Zionist regime (Israel) and other enemies of the Islamic Republic," Jahangirzadeh was quoted as saying by Iran's English-language Press TV.
"If the agency's actions lead to Iran cutting cooperation with this international body, all responsibility will be with the IAEA director general," said Jahangirzadeh, a member of parliament's national security and foreign policy committee.
The IAEA was not immediately available to comment on his allegation.
Last week, Iranian nuclear energy chief Fereydoun Abbasi-Davani said "terrorists" might have infiltrated the Vienna-based agency. He suggested the IAEA included too much sensitive information about Iran's nuclear program in its reports that he said could be used by saboteurs.
Western diplomats dismissed his allegations as an attempt to distract attention away from the agency's bid to gain access to a site in Iran it suspects was used for nuclear weapons research, something Tehran denies.
Iran blames Israel and its Western allies for the assassination of nuclear scientists in Iran, including an unsuccessful attempt on Abbasi-Davani in November 2010. It also blames those countries for computer viruses that appeared designed to damage its nuclear machinery.
The 35-nation board of the agency censured Iran earlier this month for defying international demands to curb uranium enrichment and failing to address mounting disquiet about its suspected research into atomic bombs.
The resolution prompted Iran's Parliament Speaker, Ali Larijani, to cast doubt on the benefit of Iran's membership in the Nuclear Non-Proliferation Treaty (NPT), the Tehran Times reported.
The commander of Iran's Revolutionary Guards, Mohammad Ali Jafari, told a news conference last week that Tehran would withdraw from the NPT if attacked by Israel which has increased hints it may launch air strikes on Iran's nuclear sites.
Iran's parliament does not decide matters of foreign policy and national security, which are the province of Supreme Leader Ayatollah Ali Khamenei.

TOPANGA, Calif. (AP) ? He is, and likely forever will be, best known as good old Wally Cleaver, the big brother who had to bail out a goofball sibling facing one dilemma after another on the classic TV series "Leave it to Beaver."
For the last dozen years, though, Tony Dow has been carving out a new career, as a sculptor with pieces that have shown at numerous venues, including what is arguably the world's premier art museum ? the Louvre in Paris.
This weekend, more than 30 of Dow's pieces in bronze, steel and wood go on display closer to home at the Debilzan Gallery in Laguna Beach, and they could fetch several thousand dollars each from collectors. But despite his respected reputation as a sculptor, Dow acknowledges there could be as many people at Saturday's opening reception wanting to rub shoulders with the Beav's brother as see his art.
"I think it's hard, especially with the Wally image, to be taken seriously at pretty much anything other than that," he says with a chuckle and a shake of his head.
At 67, Dow has a head of grey hair and lives with his wife, Lauren, in the wooded Southern California arts colony of Topanga Canyon.
His reputation as a sculptor reached a new height four years ago when he had one of his bronze pieces accepted at 2008's Societe Nationale des Beaux-Arts, a 150-year-old art show staged annually at the Louvre.
The modest, soft-spoken Dow is quick to point out that the work ? a distinctive abstract piece titled The Warrior ? was not placed in the museum's permanent collection. And if you went to see the show that year you would not have found it anywhere near Leonardo da Vinci's "Mona Lisa."
"But it was a show that was represented by 20-some nations, and the U.S. had 14 pieces there, and there were two sculptors, and I was one of them," Dow says between sips of mango-flavored lemonade as he relaxes on a recent hot, end-of-summer day in the living room of his home.
"So it was a big deal," he adds softly with a shy smile.
Dow doesn't complain that he's still associated with his "Leave it to Beaver" character. He loved playing Wally opposite Jerry Mathers' Beaver from 1957 to 1963, so much so that he reprised the role as an adult for a TV movie and 104 more episodes of "The New Leave it To Beaver" during much of the 1980s.
Even now, he still keeps in touch with all the old gang.
"Jerry, I talked to him just a couple days ago," he says of Mathers.
Dressed in a T-shirt, shorts and flip-flops, Dow still looks about as fit as the teenage Wally did. But you probably wouldn't recognize him as that character otherwise ? except for an occasional Wally expression or mannerism.
People who stop him while he walks through his picturesque neighborhood are more likely to ask about his dog Bodie, a dreadlocked Bergamasco Shepherd. It's in the hills of his neighborhood that Dow finds much of the rough-hewn, bark-covered wood known as burl that he carves into intricately detailed sculptures.
Dow is often described as an abstract artist, but a tour of his adjacent home studio shows numerous pieces reflecting a pop art and modern style as well.
The artist himself says he's never paid much attention to labels, although he does cite as major influences the great 19th century French sculptor Auguste Rodin and the modern British abstract artist Henry Moore.
"A lot of people say his work is Rodinish, but it's all Tony," says pop artist Stephano Sutherlin, who has staged several exhibitions of Dow's sculptures at his own gallery in Little Rock, Ark.
"He doesn't copy anybody," Sutherlin said by phone from the gallery. "He likes certain people, but it's his own work, and he's very well respected."
Dow, who painted as a youngster, began sculpting more than 30 years ago when he was still acting and directing. He planned to get serious about it when he retired from television.
"The dream was to get a barn space somewhere at the beach and have a studio," he says. But the beach turned out to be too expensive, and he and his wife instead settled in Topanga in the late 1980s.
It was 12 years ago that art took front and center in his life. Dow laughs when he remembers the day when he decided it was time to take the leap from acting to sculpting. He was up for a role in a TV show and a 28-year-old executive asked, "Have you ever done comedy before," recalls Dow, co-star of one of the most classic TV comedies in history.
"Well, I sort of looked at him and I thought, 'Hmmm, maybe it is time for me to retire. Maybe it is time to take the art seriously.'"
